Conserved elements within open reading frames of mammalian Hox genes

چکیده
A recent study in BMC Evolutionary Biology shows that many of the open reading frames in mammalian Hox genes are more conserved than expected on the basis of their protein sequence. The presence of highly conserved DNA elements is thus not confined to the noncoding DNA in neighboring regions but clearly overlaps with coding sequences. These findings support an emerging view that gene regulatory and coding sequences are likely to be more intermingled than once believed.

Picking Up the Trail of Phylogenetic Footprints
Non-coding sequence in eukaryotes encodes functionally important signals for the regulation of gene expression. Since these elements are evolutionary conserved among related taxa due to stabilizing selection they accumulate less mutations than adjacent non-functional DNA. These conserved non-coding sequences with potential regulatory activity, are known as phylogenetic footprints. They can be d...
متن کاملHox clusters as models for vertebrate genome evolution.
The surprising variation in the number of Hox clusters and the genomic architecture within vertebrate lineages, especially within the ray-finned fish, reflects a history of duplications and subsequent lineage-specific gene loss. Recent research on the evolution of conserved non-coding sequences (CNS) in Hox clusters promises to reveal interesting results for functional and phenotypic diversific...
متن کاملCombinatorial Gene Regulatory Functions Underlie Ultraconserved Elements in Drosophila
Ultraconserved elements (UCEs) are discrete genomic elements conserved across large evolutionary distances. Although UCEs have been linked to multiple facets of mammalian gene regulation their extreme evolutionary conservation remains largely unexplained. Here, we apply a computational approach to investigate this question in Drosophila, exploring the molecular functions of more than 1,500 UCEs...
متن کاملRibEx: a web server for locating riboswitches and other conserved bacterial regulatory elements
We present RibEx (riboswitch explorer), a web server capable of searching any sequence for known riboswitches as well as other predicted, but highly conserved, bacterial regulatory elements. It allows the visual inspection of the identified motifs in relation to attenuators and open reading frames (ORFs). Any of the ORF's or regulatory elements' sequence can be obtained with a click and submitt...
متن کاملEvolutionary conservation suggests a regulatory function of AUG triplets in 5′-UTRs of eukaryotic genes
By comparing sequences of human, mouse and rat orthologous genes, we show that in 5'-untranslated regions (5'-UTRs) of mammalian cDNAs but not in 3'-UTRs or coding sequences, AUG is conserved to a significantly greater extent than any of the other 63 nt triplets. This effect is likely to reflect, primarily, bona fide evolutionary conservation, rather than cDNA annotation artifacts, because the ...
